Kisspeptin is a neuropeptide that plays a fundamental role in regulating the reproductive system by directly stimulating the release of gonadotropin-releasing hormone (GnRH) from the hypothalamus. This action subsequently triggers the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary gland, which are essential for proper reproductive function.

The peptide works by binding to its receptor (GPR54) on GnRH neurons, effectively serving as the master switch that activates the entire reproductive hormone cascade. Research has shown kisspeptin’s potential in treating various reproductive disorders, including hypogonadism and infertility, while also demonstrating effects on sexual behavior, emotional processing, and metabolic regulation.

Beyond its reproductive functions, kisspeptin has garnered attention for its role in inhibiting cancer metastasis, particularly in melanoma and breast cancer, earning it the name “metastin.” Studies suggest therapeutic applications may extend to treating reproductive disorders, certain cancers, and potentially metabolic conditions, with ongoing research exploring its full clinical potential.

Kisspeptin is generally well-tolerated in clinical trials, with zero serious adverse events reported across hundreds of participants.  The most frequently documented side effects are mild, transient, and self-limiting. 

  • Injection site reactions: Occur in 10–15% of users, presenting as mild warmth, redness, or discomfort that resolves within 20–40 minutes. 

  • Transient headaches: Reported in 5–8% of participants, typically lasting 1–2 hours and linked to hormonal fluctuations. 

  • Facial flushing: Affects 3–5% of users, involving brief warmth or redness in the face and neck, resolving within 10 minutes. 

  • Other mild effects: Includes nausea, lightheadedness, and temporary mood or energy changes, which usually subside as the body adjusts.

Research and community protocols for Kisspeptin-10 typically utilize subcutaneous dosing ranges of 100–300 mcg daily, often cycled for 4–8 weeks to support hypothalamic-pituitary-gonadal (HPG) axis function. 

Standard Subcutaneous Protocols

  • Starting Dose: 100 mcg once daily or every other day to assess individual response. 

  • Standard Dose: 200–300 mcg once daily, commonly administered before bed to align with natural LH pulse timing. 

  • Advanced/Research Dose: Up to 500 mcg daily or split into twice-daily injections for individuals with suboptimal response.
